Progesterone ELISA

Background

Ridgeway Science has a 30 year pedigree in producing gold standard Progesterone ELISAs, used heavily by vets and within research institutes. Our ELISAs provide an accurate, fast measurement of an animal’s progesterone level in milk, serum or plasma. Our customers find this especially useful in studying cattle, investigating how the oestrous cycle translates into timing of insemination and ovulation. The assay can also be used to measure/test and research other species and is a very handy tool for looking at reproductive disease. We have found our ELISAs have also been used as a benchmark test to compare other new and upcoming on farm aids for heat detection, like our product P4Rapid.

About the assay…

The ELISA is a competitive indirect solid phase assay. The 96 well microplate is coated with a monoclonal antibody boasting very high specificity for progesterone. Small volumes of milk (e.g. dairy cows, goats, sheep) or serum/plasma (e.g. horses, dogs, camels) can be used as test samples. They are added in conjunction with an enzyme conjugate and left to incubate. Excess is washed off and addition of a substrate produces a colour change inversely proportional to the quantity of progesterone in the sample tested. This means a strong colour equals very low progesterone and vice versa.

Our progesterone ELISA’s are not species specific.

Milk ELISA 1.0

Ridgeway Science have been manufacturing Progesterone ELISA plates for over 30 years. Our current Milk ELISA 1.0 has been on the market for 10+ years and has been used by countless research institutes, vets and other customers. This ELISA focuses on progesterone levels typically seen in cycling cows, with a 0-50ng/ml.

We currently only supply bovine milk standards (as an add on) but can provide advice on how to make standards for other species or provide buffer standards.

Milk ELISA 2.0

We have recently re-developed and validated a new version of our Milk ELISA 2.0 for use with milk to provide a highly accurate and sensitive assay, focusing on the progesterone levels we believe are of most biological importance (0-10ng/ml).  Although the dynamic range is reduced compared with Milk ELISA 1.0, the assay sensitivity and accuracy is greater for those looking at lower levels of progesterone. We have focused on progesterone levels decreasing and increasing around oestrus and therefore developed our ELISA around this.

This ELISA can be used for very close monitoring/testing of progesterone levels to confirm or deny that the animal in question is in oestrous. Additionally, the assay can be used with any milk producing species, bearing in mind they may have different progesterone profiles.

We currently only supply bovine milk standards (as an add on) but can provide advice on how to make standards for other species or provide buffer standards.

Plasma/Serum ELISA

Our Progesterone ELISA for use with plasma or serum makes use of the same chemistry as our Milk ELISA 1.0. This assay can be used with any species and its uses can range from timing of breeding/insemination, pregnancy, diagnosis of reproductive disease.

Standards can also be supplied for this ELISA. We can provide standards for a number of species so please enquire.
